Description
AGR3 encodes a member of the protein disulfide isomerase (PDI) family, localized to the endoplasmic reticulum. It is involved in oxidative protein folding and is highly expressed in mucin-secreting epithelial cells. AGR3 is upregulated in several cancers, including breast and ovarian, and may serve as a biomarker for tumor progression.
Disease Associations
| Disease category | Pathophysiological mechanism | Genomic evidence |
|---|---|---|
| Breast cancer | Overexpression of AGR3 in estrogen receptor-positive tumors; may promote cell survival and chemoresistance | PMID: 21947068 |
| Ovarian cancer | Elevated AGR3 expression in serous ovarian carcinoma; associated with poor prognosis | PMID: 23104868 |
| Mucinous adenocarcinoma | AGR3 is a marker of mucinous differentiation in various tissues | PMID: 19074856 |

Mutation functional classification
Loss of Function (LOF)
Frameshift mutations (e.g., c.523_524insA) are predicted to cause premature truncation and loss of PDI activity.
Gain of Function (GOF)
No gain-of-function mutations have been reported for AGR3.
Dominant Negative (DN)
No dominant-negative mutations have been characterized.

Protein Summary
AGR3 is a 166-amino acid protein with a thioredoxin-like domain characteristic of PDI family members. It catalyzes disulfide bond formation and rearrangement during protein folding. AGR3 is secreted and can be detected in serum, making it a potential biomarker for mucinous tumors.
