Description
ADM (adrenomedullin) encodes a 52-amino acid peptide hormone initially isolated from human pheochromocytoma. It acts as a potent vasodilator, regulates blood pressure, promotes angiogenesis, and modulates cell growth. The gene is expressed in various tissues including adrenal medulla, heart, lung, kidney, and vascular endothelium. ADM signaling occurs through calcitonin receptor-like receptor (CALCRL) and receptor activity-modifying proteins (RAMPs).
Disease Associations
| Disease category | Pathophysiological mechanism | Genomic evidence |
|---|---|---|
| Hypertension | ADM overexpression leads to vasodilation and reduced blood pressure; polymorphisms associated with essential hypertension. | NCBI Gene, OMIM |
| Cardiovascular Disease | Elevated plasma ADM correlates with heart failure severity and prognosis; protective role in myocardial ischemia. | NCBI Gene, OMIM |
| Cancer (multiple types) | ADM promotes tumor angiogenesis and growth; upregulated in breast, lung, colorectal, and pancreatic cancers. | COSMIC, NCBI Gene |
| Sepsis | ADM levels increase in sepsis; acts as a compensatory vasodilator; potential biomarker. | NCBI Gene |
| Preeclampsia | Altered ADM expression implicated in placental vascular dysfunction. | OMIM |

Protein Summary
Adrenomedullin (P35318) is a 52-amino acid secreted peptide with an N-terminal signal peptide and a C-terminal amidation site. It contains a single disulfide bond forming a ring structure essential for receptor binding. The protein is processed from a larger precursor (proadrenomedullin) which also yields proadrenomedullin N-terminal 20 peptide (PAMP). ADM binds to the calcitonin receptor-like receptor (CALCRL) in complex with RAMP2 or RAMP3, activating Gs and leading to cAMP production. It is a potent vasodilator, inhibits apoptosis, and stimulates angiogenesis.
