Description
ADAM32 (ADAM metallopeptidase domain 32) is a member of the ADAM (a disintegrin and metalloproteinase) family. This gene encodes a transmembrane protein that contains a metalloproteinase domain, a disintegrin domain, a cysteine-rich region, and an epidermal growth factor (EGF)-like domain. ADAM32 is predominantly expressed in the testis and is thought to play a role in spermatogenesis, cell adhesion, and proteolysis. Alternative splicing results in multiple transcript variants.
Disease Associations
| Disease category | Pathophysiological mechanism | Genomic evidence |
|---|---|---|
| Lung cancer | ADAM32 mutations (e.g., missense) may alter metalloproteinase activity, potentially affecting cell migration and invasion. | COSMIC database reports somatic mutations in lung adenocarcinoma samples. |
| Colorectal cancer | Somatic missense and nonsense mutations in ADAM32 have been identified in colorectal tumors, though functional impact remains under investigation. | COSMIC database; limited functional validation. |
| Testicular germ cell tumors | Given high testis expression, ADAM32 dysregulation may contribute to germ cell tumorigenesis, but direct evidence is sparse. | NCBI Gene expression data; no definitive mechanism established. |

Mutation functional classification
Loss of Function (LOF)
Nonsense mutations (e.g., p.Arg189*) likely lead to loss of function via truncated protein.
Gain of Function (GOF)
No gain-of-function mutations have been characterized for ADAM32.
Dominant Negative (DN)
No dominant-negative mutations reported for ADAM32.

Protein Summary
ADAM32 is a transmembrane metalloproteinase of 789 amino acids (UniProt Q8TC27). It contains a signal peptide, a prodomain, a metalloproteinase domain with a zinc-binding motif (HEXXHXXGXXH), a disintegrin domain, a cysteine-rich domain, and an EGF-like domain. The protein is predicted to function in ectodomain shedding and cell-cell adhesion, particularly in the testis. Its catalytic activity has not been fully characterized experimentally.
