Description

ZFP36L2 encodes a member of the TIS11 family of CCCH tandem zinc finger proteins. The protein binds to AU-rich elements (AREs) in the 3' untranslated regions of target mRNAs, promoting their deadenylation and decay. It plays a critical role in post-transcriptional regulation of gene expression, particularly in immune response, hematopoiesis, and cell proliferation.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Acute myeloid leukemia Loss of ZFP36L2 expression leads to increased stability of oncogenic mRNAs (e.g., MYC, BCL2), promoting leukemogenesis. PMID: 24829207
B-cell lymphoma ZFP36L2 acts as a tumor suppressor by destabilizing mRNAs encoding pro-survival factors. PMID: 25691752
Inflammatory diseases Dysregulation of ZFP36L2 alters cytokine mRNA stability, contributing to chronic inflammation. PMID: 21217775

Mutation functional classification

Loss of Function (LOF)

Nonsense and frameshift mutations that truncate the protein or disrupt the CCCH zinc finger domains lead to loss of mRNA destabilization activity.

Gain of Function (GOF)

No gain-of-function mutations have been reported in ZFP36L2.

Dominant Negative (DN)

No dominant-negative mutations have been characterized for ZFP36L2.

Pathways

• mRNA surveillance pathway (Reactome: R-HSA-72163)
• AU-rich element (ARE) mediated mRNA decay (Reactome: R-HSA-450531)

Protein Summary

ZFP36L2 is a 482-amino acid protein containing two CCCH-type zinc finger domains that mediate sequence-specific binding to AU-rich elements (AREs) in target mRNAs. It recruits the CCR4-NOT deadenylase complex to promote mRNA deadenylation and decay. The protein is predominantly cytoplasmic but can shuttle to the nucleus. Post-translational modifications include phosphorylation by MAP kinases, which regulates its stability and activity.
