Description

YTHDF2 is a member of the YTH domain family of RNA-binding proteins that specifically recognize and bind N6-methyladenosine (m6A)-modified RNAs. It plays a key role in post-transcriptional regulation by promoting the degradation of m6A-containing transcripts, thereby influencing mRNA stability, gene expression, and cellular processes such as stem cell differentiation, immune response, and tumorigenesis.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Acute Myeloid Leukemia YTHDF2 overexpression promotes leukemogenesis by destabilizing m6A-modified tumor suppressor mRNAs PMID: 29507755
Hepatocellular Carcinoma YTHDF2 downregulation correlates with poor prognosis and increased cell proliferation PMID: 30401839
Colorectal Cancer YTHDF2 regulates Wnt/β-catenin signaling via m6A-dependent mRNA decay PMID: 31570779

Protein Summary

YTHDF2 is a 579-amino acid protein containing an N-terminal proline-rich region and a C-terminal YTH domain that specifically recognizes m6A-modified RNAs. It localizes to cytoplasmic foci and stress granules, where it recruits the CCR4-NOT deadenylase complex to promote deadenylation and degradation of target mRNAs. YTHDF2 is essential for embryonic stem cell self-renewal and differentiation, and its dysregulation is implicated in multiple cancers.
