Description

YBX3 (Y-Box Binding Protein 3) encodes a member of the Y-box binding protein family, which contains a cold-shock domain. This protein functions as a DNA- and RNA-binding protein, involved in transcriptional regulation, mRNA splicing, and translational control. It plays roles in cell proliferation, differentiation, and stress response. YBX3 is also known as CSDA (cold shock domain protein A) and ZONAB (zonula occludens-1 associated nucleic acid binding protein).

Mutation functional classification

Loss of Function (LOF)

Rare missense variants in the cold-shock domain (e.g., p.Arg34Cys) may impair DNA/RNA binding, reducing transcriptional activity.

Gain of Function (GOF)

No well-characterized gain-of-function mutations reported in YBX3.

Dominant Negative (DN)

Not established for YBX3.

Pathways

• Wnt signaling pathway
• mRNA surveillance pathway
• RNA transport

Protein Summary

YBX3 is a 372-amino acid protein containing a conserved cold-shock domain (CSD) at the N-terminus, which mediates nucleic acid binding. It shuttles between nucleus and cytoplasm, regulating transcription and translation. YBX3 interacts with tight junction protein ZO-1 (TJP1) and modulates cell proliferation. Overexpression in multiple cancers suggests an oncogenic role.
