Description
The VCAN gene encodes versican, a large chondroitin sulfate proteoglycan of the extracellular matrix. Versican plays key roles in cell adhesion, proliferation, migration, and tissue morphogenesis. It is involved in inflammation, cancer progression, and cardiovascular diseases.

Mutation functional classification
Loss of Function (LOF)
Nonsense and frameshift mutations lead to truncated versican, impairing extracellular matrix integrity.
Gain of Function (GOF)
Not well documented; overexpression in cancer may act as gain-of-function.
Dominant Negative (DN)
Missense mutations in GAG attachment regions may disrupt versican interactions.

Protein Summary
Versican is a large chondroitin sulfate proteoglycan that binds hyaluronan and other ECM components. It modulates cell adhesion, migration, and proliferation through interactions with CD44, integrins, and growth factors. Alternative splicing generates four isoforms (V0, V1, V2, V3) with distinct functions.
