Description

UNC5C encodes a member of the UNC-5 family of netrin receptors, which are transmembrane proteins that mediate axon guidance and cell migration in response to netrin-1. In the absence of netrin-1, UNC5C can induce apoptosis, acting as a dependence receptor. Loss of UNC5C expression is associated with colorectal cancer and other malignancies, suggesting a tumor suppressor role.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Colorectal Cancer Loss of UNC5C expression via promoter methylation leads to reduced apoptosis and increased tumor progression. PMID: 12446744, COSMIC
Breast Cancer UNC5C downregulation correlates with poor prognosis and metastasis. PMID: 21804531
Pancreatic Cancer Epigenetic silencing of UNC5C contributes to tumor growth. PMID: 19029981

Mutation functional classification

Loss of Function (LOF)

Nonsense mutations and promoter methylation lead to loss of UNC5C expression or function, impairing apoptosis and promoting tumorigenesis.

Gain of Function (GOF)

No confirmed gain-of-function mutations reported.

Dominant Negative (DN)

No dominant-negative mutations described.

Pathways

• Netrin-1 signaling (Reactome: R-HSA-373752)
• Apoptosis (KEGG: hsa04210)
• Axon guidance (KEGG: hsa04360)

Protein Summary

UNC5C is a transmembrane netrin receptor of the immunoglobulin superfamily. It contains two immunoglobulin-like domains and two thrombospondin type-1 domains in the extracellular region, and a death domain in the intracellular region. In the absence of netrin-1, UNC5C recruits caspase activators to induce apoptosis. It is widely expressed in neural tissues and at lower levels in epithelial tissues. Loss of UNC5C function is implicated in cancer progression.
