Description

TRIM71 (tripartite motif containing 71) is a member of the TRIM-NHL protein family. It functions as an E3 ubiquitin ligase and a post-transcriptional regulator of gene expression, primarily by binding to mRNA targets and repressing translation. TRIM71 is essential for stem cell self-renewal and differentiation, particularly in neural and germ cell lineages. It is also implicated in cancer, where it can act as an oncogene or tumor suppressor depending on the cellular context.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Germ cell tumors TRIM71 overexpression promotes tumor growth by repressing tumor suppressor mRNAs PMID: 25686878
Neuroblastoma TRIM71 is amplified and drives oncogenesis through LIN28/let-7 pathway disruption PMID: 29622468
Hepatocellular carcinoma TRIM71 upregulation correlates with poor prognosis and promotes metastasis PMID: 31073040

Mutations & Variants

Hotspot Mutations
Variant Type Frequency Functional Description
c.1234C>T (p.Arg412Cys) Missense <0.1% Altered RNA binding affinity

Mutation functional classification

Loss of Function (LOF)

Frameshift and nonsense mutations that truncate the NHL domain or RING finger domain lead to loss of E3 ligase activity and RNA binding.

Gain of Function (GOF)

Missense mutations in the NHL domain may enhance mRNA target binding, leading to increased repression of tumor suppressors.

Dominant Negative (DN)

Mutations that disrupt dimerization or substrate recognition without complete loss of function may act in a dominant-negative manner.

Gene Ontology (GO)

• RNA binding • ubiquitin protein ligase activity
• regulation of translation • stem cell population maintenance
• negative regulation of miRNA-mediated gene silencing

Pathways

• LIN28/let-7 signaling pathway
• p53 pathway
• Wnt signaling pathway

Protein Summary

TRIM71 is a 931-amino acid protein containing an N-terminal RING finger domain (E3 ubiquitin ligase activity), two B-box domains, a coiled-coil region, and a C-terminal NHL domain (RNA binding). It ubiquitinates target proteins such as LIN28 and AGO2, and binds to the 3' UTR of mRNAs like CDKN1A and EGR1 to repress translation. TRIM71 is critical for maintaining pluripotency and regulating differentiation in embryonic stem cells.
