Description

TRIM69 (Tripartite Motif Containing 69) is a protein-coding gene that belongs to the tripartite motif (TRIM) family. The encoded protein contains a RING finger domain, B-box type 1 and type 2 domains, and a coiled-coil region. TRIM69 functions as an E3 ubiquitin ligase and plays a role in antiviral innate immunity by restricting viral replication, particularly against flaviviruses such as dengue virus. It also has been implicated in cell cycle regulation and may act as a tumor suppressor in certain cancers.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Dengue virus infection TRIM69 restricts dengue virus replication by ubiquitinating viral NS3 protein PMID: 29386363
Hepatocellular carcinoma Downregulation of TRIM69 correlates with poor prognosis; loss promotes proliferation and metastasis PMID: 31827240
Breast cancer Reduced TRIM69 expression associated with aggressive tumor features and shorter survival PMID: 32644122

Mutation functional classification

Loss of Function (LOF)

Frameshift mutations (e.g., c.1345_1346insA) are predicted to cause premature truncation and loss of E3 ubiquitin ligase activity.

Gain of Function (GOF)

No gain-of-function mutations have been reported for TRIM69.

Dominant Negative (DN)

No dominant-negative mutations have been characterized.

Pathways

• Ubiquitin mediated proteolysis (KEGG: hsa04120)
• RIG-I-like receptor signaling pathway (KEGG: hsa04622)
• Antiviral mechanism by TRIM proteins (Reactome: R-HSA-936440)

Protein Summary

TRIM69 is a 497-amino acid protein with a molecular weight of approximately 56 kDa. It contains an N-terminal RING finger domain (residues 15-55) that confers E3 ubiquitin ligase activity, followed by two B-box domains (B-box1: 91-133; B-box2: 147-189) and a coiled-coil region (residues 200-250). The protein localizes to the cytoplasm and is involved in ubiquitination of viral proteins, thereby restricting viral replication. TRIM69 also interacts with cellular proteins to modulate cell cycle progression and apoptosis.
