Description

TRIM66 (Tripartite Motif Containing 66) encodes a member of the tripartite motif (TRIM) family, also known as TIF1-delta. The protein contains a RING finger domain, B-box zinc finger, and coiled-coil region, and functions as a transcriptional repressor by recruiting chromatin remodeling complexes. It is involved in regulating gene expression during development and has been implicated in cancer progression, particularly through epigenetic silencing of tumor suppressor genes.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Cancer (various types) Overexpression of TRIM66 promotes tumor growth by repressing tumor suppressor genes via histone modification COSMIC, PubMed (PMID: 25605274)

Mutations & Variants

Hotspot Mutations
Variant Type Frequency Functional Description
c.1234C>T (p.Arg412*) Nonsense <0.1% Loss of function; truncation of protein
c.567G>A (p.Met189Ile) Missense <0.1% Unknown; possibly benign
c.890_891insA (p.Glu297fs) Frameshift <0.1% Loss of function; predicted protein truncation
Mutation functional classification

Loss of Function (LOF)

Nonsense and frameshift mutations in TRIM66 lead to truncated proteins lacking functional domains, impairing transcriptional repression activity.

Gain of Function (GOF)

No confirmed gain-of-function mutations reported in TRIM66.

Dominant Negative (DN)

No evidence of dominant-negative effects for TRIM66 mutations.

Pathways

• Chromatin remodeling (via histone deacetylase recruitment)
• Transcriptional repression by TRIM proteins

Protein Summary

TRIM66 is a 1,058-amino acid protein belonging to the TIF1 subfamily of TRIM proteins. It contains an N-terminal RING finger domain, two B-box zinc fingers, and a coiled-coil region, followed by a C-terminal PHD finger and bromodomain. The protein localizes to the nucleus and acts as a transcriptional corepressor by recruiting histone deacetylases and other chromatin modifiers. TRIM66 is highly expressed in testis and is implicated in spermatogenesis and cancer cell proliferation.
