Description

TRIM60 (tripartite motif containing 60) is a protein-coding gene belonging to the tripartite motif (TRIM) family, characterized by a RING finger domain, B-box zinc finger, and coiled-coil region. TRIM60 is involved in innate immune signaling, particularly as a negative regulator of antiviral responses. It is expressed in multiple tissues and has been implicated in certain cancers through altered expression and potential oncogenic roles.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Cancer (various types) Altered TRIM60 expression may modulate NF-κB and interferon pathways, affecting tumor progression Limited; expression changes observed in cancer cell lines and tumor samples (COSMIC)
Viral infections TRIM60 negatively regulates RIG-I-like receptor signaling, reducing antiviral cytokine production Inferred from functional studies (UniProt)

Mutation functional classification

Loss of Function (LOF)

Frameshift mutations (e.g., p.Thr278Asnfs*5) are predicted to cause premature truncation and loss of TRIM60 protein function.

Gain of Function (GOF)

No gain-of-function mutations have been reported for TRIM60.

Dominant Negative (DN)

No dominant-negative mutations have been described for TRIM60.

Pathways

• RIG-I-like receptor signaling pathway (negative regulation)
• Innate immune system

Protein Summary

TRIM60 is a 493-amino acid protein with a molecular weight of approximately 56 kDa. It contains a RING-type zinc finger domain (residues 15-55) that confers E3 ubiquitin ligase activity, two B-box zinc fingers (residues 88-131 and 140-183), and a coiled-coil region (residues 197-240). TRIM60 localizes to the cytoplasm and negatively regulates antiviral signaling by targeting RIG-I for ubiquitination and degradation. Its expression is highest in testis and spleen, with lower levels in other tissues.
