Description

TRIM27 (tripartite motif containing 27), also known as RFP (ret finger protein), is a member of the tripartite motif (TRIM) family. It contains a RING finger domain, B-box type 1 and type 2, and a coiled-coil region. TRIM27 functions as an E3 ubiquitin ligase and is involved in regulating innate immune signaling, apoptosis, and transcriptional repression. It is frequently overexpressed in various cancers and has been implicated in the pathogenesis of several diseases.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Cancer (multiple types) Overexpression of TRIM27 promotes cell proliferation, inhibits apoptosis, and enhances tumor growth through modulation of NF-κB and PI3K/AKT pathways. Multiple studies (e.g., PMID: 25642768, PMID: 28431248)
Inflammatory diseases TRIM27 negatively regulates TLR4-mediated NF-κB activation and pro-inflammatory cytokine production. PMID: 22607943
Autoimmune disorders TRIM27 modulates type I interferon production and may contribute to autoimmune pathogenesis. PMID: 22607943

Mutations & Variants

Hotspot Mutations
Variant Type Frequency Functional Description
c.1A>G (p.Met1?) Missense Rare Potential loss of start codon; functional impact unknown
c.100C>T (p.Arg34Trp) Missense Rare Located in RING domain; may affect E3 ligase activity
c.200G>A (p.Arg67Gln) Missense Rare Located in B-box domain; functional significance unclear
Mutation functional classification

Loss of Function (LOF)

Mutations in the RING finger domain may impair E3 ubiquitin ligase activity, leading to reduced ubiquitination of substrates such as NEMO and IκBα, thereby affecting NF-κB signaling.

Gain of Function (GOF)

Overexpression of wild-type TRIM27 in cancers suggests a gain-of-function oncogenic role, but specific gain-of-function mutations have not been well characterized.

Dominant Negative (DN)

No dominant-negative mutations have been reported for TRIM27.

Pathways

• NF-kappa B signaling pathway
• PI3K-Akt signaling pathway
• Innate immune system
• Ubiquitin mediated proteolysis

Protein Summary

TRIM27 is a 513-amino acid protein with a molecular weight of approximately 57 kDa. It contains an N-terminal RING finger domain (required for E3 ubiquitin ligase activity), two B-box zinc finger domains, a coiled-coil region, and a C-terminal B30.2/SPRY domain. The protein localizes to both the cytoplasm and nucleus. As an E3 ubiquitin ligase, TRIM27 ubiquitinates substrates such as NEMO and IκBα, leading to their degradation and modulation of NF-κB signaling. It also interacts with other proteins including BAF53 and is involved in transcriptional repression. TRIM27 is overexpressed in several cancers and is considered a potential oncogene.
