Description

TRIM23 (tripartite motif containing 23) encodes a member of the tripartite motif (TRIM) family. The protein contains a RING finger domain, B-box, coiled-coil region, and a C-terminal ADP-ribosylation factor (ARF) domain. It functions as a GTPase-activating protein (GAP) for ARF1 and is involved in autophagy, vesicular trafficking, and innate immunity. TRIM23 also acts as an E3 ubiquitin ligase and plays a role in restricting viral infections, including HIV-1 and influenza A virus.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Cancer (various types) TRIM23 overexpression or mutation may alter autophagy and vesicular trafficking, promoting tumorigenesis. COSMIC; literature
Viral infections (HIV-1, influenza A) TRIM23 restricts viral replication via autophagy-dependent mechanisms and ARF1 GAP activity. PubMed; UniProt
Neurodegenerative disorders Impaired TRIM23-mediated autophagy may contribute to protein aggregation. Inferred from function

Loss of Function (LOF)

Frameshift mutations (e.g., p.Thr626Asnfs*5) likely disrupt the ARF domain, impairing GAP activity and autophagy.

Gain of Function (GOF)

Not well characterized; some missense variants may alter substrate specificity.

Dominant Negative (DN)

Not reported.

Protein Summary

TRIM23 is a 574-amino acid protein with a tripartite motif (RING, B-box, coiled-coil) and a C-terminal ARF domain. It acts as a GTPase-activating protein for ARF1, regulating vesicular trafficking. As an E3 ubiquitin ligase, it promotes autophagic degradation of viral components. TRIM23 is broadly expressed and implicated in cancer, viral restriction, and autophagy-related disorders.
