Description

TP53I3 (tumor protein p53 inducible protein 3) is a protein-coding gene that encodes a pro-apoptotic protein induced by the tumor suppressor p53 in response to DNA damage. The protein localizes to the cytoplasm and nucleus and is involved in the generation of reactive oxygen species (ROS) and the induction of apoptosis. TP53I3 is also known as PIG3 (p53-induced gene 3) and plays a role in the cellular response to genotoxic stress.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Breast cancer TP53I3 expression is regulated by p53; loss of p53 function leads to reduced TP53I3 expression and impaired apoptosis. NCBI Gene, PubMed: 10652319
Colorectal cancer TP53I3 is upregulated in response to DNA damage; mutations in p53 pathway may alter TP53I3 expression. NCBI Gene, PubMed: 10652319
Lung cancer TP53I3 expression is associated with p53 status; potential role in tumor suppression. NCBI Gene, PubMed: 10652319

Loss of Function (LOF)

Mutations that disrupt the pro-apoptotic function of TP53I3 may impair p53-mediated apoptosis.

Gain of Function (GOF)

No known gain-of-function mutations reported.

Dominant Negative (DN)

No known dominant-negative mutations reported.

Protein Summary

TP53I3 encodes a 232-amino acid protein with a molecular weight of approximately 26 kDa. The protein contains a putative NAD(P)H quinone oxidoreductase domain and is involved in the generation of reactive oxygen species (ROS) leading to apoptosis. It is induced by p53 in response to DNA damage and localizes to both the cytoplasm and nucleus.
