Description

TNFRSF10D encodes a member of the TNF-receptor superfamily, known as decoy receptor 2 (DcR2) or TRAIL receptor 4. This receptor contains a truncated cytoplasmic death domain and does not induce apoptosis upon ligand binding. It acts as an antagonistic decoy receptor for TRAIL (TNFSF10), inhibiting TRAIL-mediated apoptosis by competing with death receptors DR4 and DR5. The gene is located on chromosome 8p21.3 and is implicated in cancer cell resistance to TRAIL-based therapies.

Protein Summary

TNFRSF10D (DcR2) is a 386-amino acid type I transmembrane protein with two extracellular cysteine-rich domains and a truncated cytoplasmic death domain. It lacks the functional death domain required for apoptosis induction. DcR2 binds TRAIL (TNFSF10) with high affinity but does not transmit a death signal, thereby acting as a decoy receptor that protects cells from TRAIL-mediated apoptosis. The protein is expressed at low levels in normal tissues but is upregulated in various cancers, contributing to immune evasion and therapy resistance.
