Description
TNFRSF10C encodes a member of the TNF-receptor superfamily, known as decoy receptor 1 (DcR1) or TRAIL-R3. This receptor contains extracellular TRAIL-binding domains but lacks a functional cytoplasmic death domain, thus it cannot induce apoptosis. It acts as an antagonistic decoy receptor that competes with death receptors DR4 (TNFRSF10A) and DR5 (TNFRSF10B) for TRAIL binding, thereby inhibiting TRAIL-mediated apoptosis. DcR1 is a glycosylphosphatidylinositol (GPI)-anchored cell surface protein. Overexpression of DcR1 has been observed in various cancers and may contribute to tumor resistance to TRAIL-based therapies.

Protein Summary
TNFRSF10C (DcR1) is a 259-amino acid GPI-anchored cell surface protein. It contains two extracellular cysteine-rich domains (CRDs) that bind TRAIL with high affinity but lacks an intracellular death domain. DcR1 functions as a decoy receptor, competing with DR4 and DR5 for TRAIL binding and thereby inhibiting apoptosis. It is expressed in many tissues but at very low levels; overexpression in tumors contributes to TRAIL resistance. The protein is encoded by the TNFRSF10C gene located on chromosome 8p21.3.
