Description

TFPI2 (Tissue Factor Pathway Inhibitor 2) encodes a serine protease inhibitor belonging to the Kunitz-type inhibitor family. The protein inhibits a broad range of serine proteases including plasmin, trypsin, chymotrypsin, and factor VIIa/tissue factor complex. It is highly expressed in the placenta and is involved in extracellular matrix remodeling, coagulation regulation, and tumor suppression. Epigenetic silencing via promoter methylation is frequently observed in various cancers.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Placental dysfunction Altered TFPI2 expression may affect trophoblast invasion and coagulation balance at the maternal-fetal interface. PMID: 12368206
Colorectal cancer Promoter hypermethylation of TFPI2 leads to transcriptional silencing and loss of tumor suppressor function. PMID: 17638982
Gastric cancer Reduced TFPI2 expression due to methylation correlates with tumor progression and poor prognosis. PMID: 20097252
Pancreatic cancer TFPI2 methylation is a frequent epigenetic event associated with invasive phenotype. PMID: 15696243
Hepatocellular carcinoma TFPI2 downregulation via methylation promotes metastasis and angiogenesis. PMID: 20629109

Protein Summary

TFPI2 is a 213-amino-acid secreted protein containing three Kunitz domains. It inhibits serine proteases such as plasmin, trypsin, and the factor VIIa-tissue factor complex. The protein is heavily glycosylated and localizes to the extracellular matrix. It plays a key role in regulating pericellular proteolysis, coagulation, and tumor invasion. Loss of TFPI2 expression via promoter methylation is a common event in multiple cancers, contributing to metastatic progression.
