Description

The TEC gene encodes a non-receptor tyrosine kinase belonging to the Tec family, which also includes BTK, ITK, TXK, and BMX. Tec kinase is predominantly expressed in hematopoietic cells and plays a critical role in signal transduction from antigen receptors, cytokine receptors, and integrins. It is involved in T-cell activation, B-cell development, and mast cell degranulation. Mutations and dysregulation of TEC are associated with immunodeficiency and hematological malignancies, making it a therapeutic target for kinase inhibitors.

Protein Summary

Tec is a 631-amino acid cytoplasmic tyrosine kinase with a PH domain, SH3 domain, SH2 domain, and a C-terminal kinase domain. It is activated by Src family kinases and PI3K products. Tec phosphorylates downstream targets such as PLCγ, VAV, and STAT3, regulating calcium flux, cytoskeletal rearrangement, and gene expression. Its expression is restricted to hematopoietic tissues, and it is essential for normal immune function.
