Description
TAF7 (TATA-box binding protein associated factor 7) encodes a component of the transcription factor IID (TFIID) complex. This protein is essential for transcription initiation by RNA polymerase II, interacting with TBP and other TAFs to facilitate promoter recognition and pre-initiation complex assembly. TAF7 also plays roles in cell cycle progression and DNA repair.

Mutation functional classification
Loss of Function (LOF)
Frameshift mutations (e.g., p.Thr230Asnfs*2) likely result in truncated protein and loss of TFIID complex interaction.
Gain of Function (GOF)
No well-characterized gain-of-function mutations reported.
Dominant Negative (DN)
Missense mutations in conserved domains may disrupt TFIID assembly, potentially acting in a dominant-negative manner.

Protein Summary
TAF7 is a 55 kDa protein that contains a histone fold domain and a TBP-binding region. It is a stable component of the TFIID complex, where it helps recruit RNA polymerase II to core promoters. TAF7 also interacts with transcription factors such as Sp1 and p53, modulating their activity. Post-translational modifications include phosphorylation, which may regulate its function during the cell cycle.
