Description

TACC2 (transforming acidic coiled-coil containing protein 2) encodes a member of the transforming acidic coiled-coil (TACC) family. The protein localizes to centrosomes and microtubules during interphase and to the mitotic spindle during mitosis. It plays a role in centrosome integrity, microtubule stabilization, and cell cycle progression. TACC2 is involved in transcriptional regulation and has been implicated in tumor suppression and oncogenesis depending on cellular context.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Breast cancer Altered TACC2 expression affects centrosome function and genomic stability; loss of expression correlates with tumor progression PMID: 15064718, COSMIC
Glioma TACC2 amplification or overexpression contributes to mitotic spindle defects and aneuploidy PMID: 24803665, COSMIC
Lung cancer Downregulation of TACC2 is associated with poor prognosis and increased metastasis PMID: 21804532
Prostate cancer TACC2 acts as a putative tumor suppressor; reduced expression linked to aggressive disease PMID: 15604208

Mutation functional classification

Loss of Function (LOF)

Nonsense and frameshift mutations (e.g., p.Arg412*, p.Glu190fs) lead to truncated protein lacking C-terminal TACC domain, impairing centrosomal localization and microtubule binding.

Gain of Function (GOF)

Not clearly established; amplification of TACC2 in some cancers may lead to overexpression and aberrant mitotic spindle function.

Dominant Negative (DN)

Not reported for TACC2.

Pathways

• Centrosome maturation and spindle assembly
• Microtubule cytoskeleton organization
• Cell cycle
• mitotic

Protein Summary

TACC2 is a 639-amino-acid protein (isoform 1) containing an N-terminal acidic region and a C-terminal coiled-coil TACC domain. It localizes to centrosomes and microtubules, interacting with ch-TOG (CKAP5) and clathrin to stabilize microtubules during mitosis. TACC2 is involved in centrosome integrity, spindle assembly, and cell cycle progression. Alternative splicing generates multiple isoforms with distinct expression patterns and functions.
