Description

STX11 encodes syntaxin 11, a member of the syntaxin family of SNARE proteins involved in vesicle docking and fusion. Syntaxin 11 is predominantly expressed in immune cells and is essential for cytotoxic granule exocytosis in natural killer (NK) cells and cytotoxic T lymphocytes (CTLs). Mutations in STX11 cause familial hemophagocytic lymphohistiocytosis type 4 (FHL4), a severe hyperinflammatory disorder characterized by impaired lymphocyte cytotoxicity.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Familial hemophagocytic lymphohistiocytosis type 4 (FHL4) Loss-of-function mutations in STX11 disrupt SNARE complex formation, preventing cytotoxic granule fusion and release, leading to uncontrolled immune activation and hypercytokinemia. OMIM #603552; ClinVar; NCBI Gene
Hemophagocytic lymphohistiocytosis (HLH) STX11 mutations impair perforin/granzyme secretion from cytotoxic cells, resulting in defective clearance of infected or malignant cells and persistent immune stimulation. PubMed; OMIM

Mutations & Variants

Hotspot Mutations
Variant Type Frequency Functional Description
c.174G>A (p.Trp58*) Nonsense Rare (found in FHL4 families) Loss of function; premature truncation of syntaxin 11
c.328C>T (p.Arg110*) Nonsense Rare Loss of function; truncated protein
c.401delG (p.Gly134Alafs*15) Frameshift Rare Loss of function; frameshift leads to early stop codon
c.527G>A (p.Arg176Gln) Missense Rare Loss of function; disrupts SNARE binding
Mutation functional classification

Loss of Function (LOF)

Most STX11 mutations are loss-of-function, leading to defective cytotoxic granule exocytosis and FHL4.

Gain of Function (GOF)

No gain-of-function mutations reported.

Dominant Negative (DN)

Not described; FHL4 is autosomal recessive.

Pathways

• Reactome: R-HSA-202733 - Cell surface interactions at the vascular wall

Protein Summary

Syntaxin 11 is a 287-amino-acid protein (32 kDa) belonging to the syntaxin family of t-SNAREs. It contains a C-terminal transmembrane domain and a SNARE motif that mediates interactions with other SNARE proteins (e.g., VAMP8, SNAP23) to facilitate membrane fusion. In cytotoxic lymphocytes, syntaxin 11 is essential for the fusion of cytotoxic granules with the plasma membrane, enabling the release of perforin and granzymes to kill target cells. Loss of syntaxin 11 function leads to familial hemophagocytic lymphohistiocytosis type 4 (FHL4).
