Description
STMN1 encodes stathmin 1, a cytosolic phosphoprotein that regulates microtubule dynamics by sequestering tubulin dimers and promoting microtubule catastrophe. It is involved in cell cycle progression, signal transduction, and cellular differentiation. Overexpression is observed in various cancers and correlates with poor prognosis.
Disease Associations
| Disease category | Pathophysiological mechanism | Genomic evidence |
|---|---|---|
| Acute myeloid leukemia | Overexpression of STMN1 disrupts microtubule stability, promoting proliferation and resistance to chemotherapy. | PMID: 21551233 |
| Breast carcinoma | High STMN1 expression associated with aggressive tumor phenotype and poor survival. | PMID: 24840125 |
| Non-small cell lung cancer | STMN1 upregulation linked to increased metastasis and chemoresistance. | PMID: 26967287 |

Protein Summary
Stathmin 1 (STMN1) is a 149-amino-acid phosphoprotein that acts as a key regulator of microtubule dynamics. It binds tubulin heterodimers, preventing their polymerization and promoting microtubule catastrophe. Phosphorylation by various kinases (e.g., CDKs, PKA, MAPK) inactivates its microtubule-destabilizing activity, allowing cell cycle progression. STMN1 is highly expressed in the brain and in many cancers, where it contributes to proliferation, invasion, and drug resistance.
