Description

STK38 (Serine/Threonine Kinase 38), also known as NDR1, is a member of the NDR (Nuclear Dbf2-Related) kinase subfamily within the AGC kinase group. It plays critical roles in cell cycle progression, centrosome duplication, apoptosis, and the Hippo signaling pathway. STK38 is activated by MST1/MST2 kinases and MOB1 co-activators, and its dysregulation is implicated in various cancers and developmental disorders.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Cancer (various types) Dysregulation of STK38 affects Hippo pathway-mediated cell proliferation and apoptosis; altered expression linked to tumorigenesis. COSMIC; literature review
Neurodevelopmental disorders Mutations in STK38 may impair neuronal cell cycle control and survival. ClinVar; limited case reports
Immune system disorders STK38 involvement in T-cell apoptosis and immune regulation. UniProt; PubMed

Protein Summary

STK38 encodes a 465-amino acid serine/threonine kinase (NDR1) with an N-terminal regulatory domain, a catalytic kinase domain, and a C-terminal extension. It is activated by phosphorylation at Thr444 and Ser281 via MST1/2 and MOB1. STK38 regulates centrosome duplication, mitotic exit, and apoptosis. It is ubiquitously expressed with highest levels in brain and testis. Mutations are rare but may contribute to cancer and developmental phenotypes.
