Description

SRPK2 (SRSF protein kinase 2) encodes a serine/arginine-rich protein kinase that phosphorylates serine/arginine-rich (SR) splicing factors, regulating pre-mRNA splicing. The kinase is involved in cell cycle progression, apoptosis, and nuclear organization. Overexpression and altered activity of SRPK2 are implicated in various cancers, including hepatocellular carcinoma, breast cancer, and leukemia, as well as in neurodegenerative disorders.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Hepatocellular carcinoma SRPK2 overexpression promotes splicing of oncogenic isoforms (e.g., VEGF, BCL-X) and enhances cell proliferation PMID: 23431147; COSMIC overexpression data
Breast cancer SRPK2 upregulation correlates with poor prognosis; phosphorylates SRSF1 to drive alternative splicing of tumor suppressor genes PMID: 25605248; ClinVar somatic variants
Acute myeloid leukemia SRPK2 fusion transcripts and kinase activation contribute to leukemogenesis via aberrant splicing of RUNX1 PMID: 27693307; COSMIC fusion data
Alzheimer's disease SRPK2-mediated phosphorylation of tau and splicing dysregulation linked to neurodegeneration PMID: 22121218; UniProt annotation

Loss of Function (LOF)

No well-characterized loss-of-function mutations reported in SRPK2; most variants are missense with uncertain impact.

Gain of Function (GOF)

Overexpression and activating mutations (e.g., p.Arg396Trp) are associated with increased kinase activity and oncogenic splicing.

Dominant Negative (DN)

No dominant-negative mutations described for SRPK2.

Pathways

• SRP-dependent cotranslational protein targeting to membrane (Reactome: R-HSA-1799339)
• Signaling by Receptor Tyrosine Kinases (Reactome: R-HSA-9006934)

Protein Summary

SRPK2 is a 688-amino-acid serine/arginine-rich protein kinase that localizes to both nucleus and cytoplasm. It specifically phosphorylates SR splicing factors (e.g., SRSF1, SRSF2) at serine residues in RS domains, modulating their nuclear import and spliceosome assembly. The protein contains a bipartite nuclear localization signal and a kinase domain. SRPK2 activity is regulated by cell cycle-dependent phosphorylation and interaction with molecular chaperones. Dysregulation of SRPK2 contributes to aberrant splicing in cancer and neurodegeneration.
