Description

The SFN gene encodes stratifin, a member of the 14-3-3 family of proteins. Stratifin (14-3-3 sigma) is a cell cycle regulator that binds to and inhibits CDC25 phosphatases, leading to G2/M arrest. It also plays roles in DNA damage response, apoptosis, and cell adhesion. SFN is frequently silenced by hypermethylation in various cancers, acting as a tumor suppressor.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Breast Cancer Loss of SFN expression via promoter hypermethylation contributes to tumor progression and poor prognosis. PMID: 10655551, COSMIC
Lung Cancer SFN downregulation is associated with increased proliferation and metastasis. PMID: 15173019, COSMIC
Gastric Cancer SFN methylation is a frequent event and correlates with advanced stage. PMID: 16189554, ClinVar
Prostate Cancer Reduced SFN expression promotes androgen-independent growth. PMID: 16951256, COSMIC
Bladder Cancer SFN hypermethylation is linked to invasive phenotype. PMID: 15510170, ClinVar

Loss of Function (LOF)

Promoter hypermethylation leading to transcriptional silencing is the primary loss-of-function mechanism in cancer.

Gain of Function (GOF)

No recurrent gain-of-function mutations reported.

Dominant Negative (DN)

No dominant-negative mutations described.

Pathways

• Cell Cycle
• G2/M DNA damage checkpoint
• 14-3-3-mediated signaling
• p53-dependent G1/S DNA damage checkpoint

Protein Summary

Stratifin (14-3-3 sigma) is a 248-amino acid protein that functions as a homodimeric adapter. It binds phosphorylated serine/threonine residues on target proteins, modulating their activity, localization, and stability. Key targets include CDC25B, CDC25C, CDKN1A (p21), and BAX. Stratifin is essential for G2/M arrest after DNA damage and is a downstream effector of p53. Loss of SFN expression is a common event in epithelial cancers.
