Description

SEMG1 encodes semenogelin 1, the predominant protein in human semen. It is secreted by the seminal vesicles and forms a coagulum that entraps spermatozoa. Subsequent proteolysis by prostate-specific antigen (PSA) liquefies the semen, releasing motile sperm. SEMG1 also exhibits antimicrobial activity and is implicated in male infertility and prostate cancer.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Male infertility Reduced or absent SEMG1 expression impairs semen coagulation and sperm motility ClinVar, PubMed
Prostate cancer Altered SEMG1 expression and proteolysis by PSA may influence tumor progression COSMIC, PubMed

Protein Summary

Semenogelin 1 is a 462-amino acid protein secreted by seminal vesicles. It forms a gel matrix that traps spermatozoa. Cleavage by PSA (kallikrein-3) at specific tyrosine residues leads to semen liquefaction. The protein also contains fibronectin type II domains and has antimicrobial properties.
