Description
SCAND1 (SCAN domain-containing 1) encodes a protein that belongs to the SCAN domain family of zinc finger transcription factors. The SCAND1 protein contains a conserved SCAN domain at the N-terminus and a C2H2-type zinc finger motif, enabling DNA binding and transcriptional regulation. It is involved in cell proliferation, differentiation, and has been implicated in various cancers. SCAND1 can form heterodimers with other SCAN domain proteins, modulating their transcriptional activity.

Mutation functional classification
Loss of Function (LOF)
Start loss mutations (e.g., p.Met1Val) are predicted to abolish protein translation, leading to loss of SCAND1 function.
Gain of Function (GOF)
No confirmed gain-of-function mutations reported in SCAND1.
Dominant Negative (DN)
No evidence for dominant-negative effects in SCAND1.

Protein Summary
SCAND1 is a 510-amino acid protein with a molecular weight of approximately 56 kDa. It contains an N-terminal SCAN domain (residues 1-70) and a C-terminal C2H2-type zinc finger domain (residues 430-458). The SCAN domain mediates protein-protein interactions, particularly homodimerization and heterodimerization with other SCAN family members, while the zinc finger domain binds to specific DNA sequences to regulate gene expression. SCAND1 is predominantly nuclear and acts as a transcriptional activator. Post-translational modifications include phosphorylation, which may modulate its activity.
