Description

RPL34 encodes ribosomal protein L34, a component of the 60S large ribosomal subunit. This protein is involved in the initiation and elongation phases of protein synthesis. RPL34 belongs to the L34E family of ribosomal proteins and is located in the cytoplasm. The gene is highly conserved across eukaryotes. Overexpression or mutations of RPL34 have been linked to various cancers, including colorectal, gastric, and hepatocellular carcinoma, suggesting a role beyond canonical translation.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Colorectal Cancer Overexpression of RPL34 promotes cell proliferation and invasion via upregulation of cyclin D1 and MMP-9. PMID: 27557510
Gastric Cancer RPL34 is upregulated and correlates with poor prognosis; knockdown reduces cell growth and induces apoptosis. PMID: 29393407
Hepatocellular Carcinoma RPL34 overexpression enhances tumor growth and metastasis through PI3K/AKT signaling. PMID: 30343334
Ribosomopathy (general) Mutations in ribosomal proteins can cause tissue-specific disorders; RPL34 variants are rare but may contribute to Diamond-Blackfan anemia-like phenotypes. PMID: 24675556

Mutations & Variants

Hotspot Mutations
Variant Type Frequency Functional Description
c.95A>G (p.Gln32Arg) Missense <0.01% Unknown; predicted benign by SIFT
c.124C>T (p.Arg42Cys) Missense <0.01% Unknown; predicted damaging by PolyPhen-2

Mutation functional classification

Loss of Function (LOF)

Frameshift mutations (e.g., c.201_202insA) lead to premature stop codons and truncated protein, likely causing haploinsufficiency.

Gain of Function (GOF)

Not well documented; overexpression in cancers suggests possible gain-of-function through increased translation of oncogenic mRNAs.

Dominant Negative (DN)

No evidence currently available for dominant-negative effects of RPL34 mutations.

Pathways

Protein Summary

Ribosomal protein L34 (RPL34) is a 117-amino-acid protein with a molecular weight of approximately 13.5 kDa. It is a component of the 60S large ribosomal subunit, where it contributes to the structural integrity and function of the ribosome. The protein contains a conserved L34E domain. RPL34 interacts with rRNA and other ribosomal proteins to facilitate peptide bond formation during translation. Beyond its canonical role, RPL34 has been implicated in cell cycle regulation, apoptosis, and tumorigenesis, likely through selective translation of specific mRNAs.
