Description
PTPN3 encodes a member of the protein tyrosine phosphatase (PTP) family. PTPs are signaling molecules that regulate a variety of cellular processes including cell growth, differentiation, mitotic cycle, and oncogenic transformation. This PTP contains a C-terminal catalytic domain and an N-terminal domain with homology to the band 4.1 superfamily, which may localize the protein to the cytoskeleton. Alternative splicing results in multiple transcript variants.
Disease Associations
| Disease category | Pathophysiological mechanism | Genomic evidence |
|---|---|---|
| Colorectal cancer | Loss of PTPN3 expression may promote tumorigenesis through altered MAPK signaling | PMID: 22198284 |
| Hepatocellular carcinoma | PTPN3 mutations may affect JNK signaling and cell proliferation | PMID: 24803677 |
| Breast cancer | Reduced PTPN3 expression correlates with poor prognosis | PMID: 25485619 |

Protein Summary
PTPN3 is a 913-amino acid protein with a molecular weight of approximately 104 kDa. It contains an N-terminal FERM domain (band 4.1, ezrin, radixin, moesin) and a C-terminal protein tyrosine phosphatase catalytic domain. The FERM domain mediates interaction with the cytoskeleton, while the phosphatase domain dephosphorylates tyrosine residues on target proteins, modulating signaling pathways such as MAPK and JNK.
