Description

PRMT6 encodes a type I protein arginine methyltransferase that catalyzes the formation of asymmetric dimethylarginine (ADMA) on histone and non-histone proteins. It is involved in transcriptional repression, RNA processing, DNA damage response, and viral replication. PRMT6 is overexpressed in several cancers and has been implicated in tumorigenesis and poor prognosis.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Breast cancer PRMT6 overexpression promotes cell proliferation and migration via methylation of histone H3R2 and non-histone targets such as NF-κB. PMID: 25977310
Prostate cancer PRMT6 enhances androgen receptor signaling and tumor growth through arginine methylation of histone H4R3. PMID: 31073040
HIV-1 infection PRMT6 methylates HIV-1 Tat protein, reducing its transactivation activity and inhibiting viral replication. PMID: 16912299
Lung cancer PRMT6 upregulation correlates with poor survival and promotes epithelial-mesenchymal transition (EMT). PMID: 31570863

Mutation functional classification

Loss of Function (LOF)

Frameshift mutations (e.g., p.Thr339Asnfs*2) likely result in truncated, non-functional protein.

Gain of Function (GOF)

No confirmed gain-of-function mutations reported in PRMT6.

Dominant Negative (DN)

No evidence of dominant-negative effects for PRMT6 mutations.

Pathways

• Arginine and proline metabolism
• Transcriptional misregulation in cancer
• p53 signaling pathway (via methylation of p53)

Protein Summary

PRMT6 is a 375-amino acid protein (42 kDa) that belongs to the protein arginine methyltransferase family. It contains a conserved methyltransferase domain and specifically generates asymmetric dimethylarginine residues. PRMT6 methylates histone H3 at arginine 2 (H3R2me2a), leading to transcriptional repression, and also modifies non-histone proteins such as p53, NF-κB, and HIV-1 Tat. Its activity is implicated in cell cycle regulation, DNA repair, and viral restriction.
