Description

PRMT2 (Protein Arginine Methyltransferase 2) encodes a type I arginine methyltransferase that catalyzes the formation of asymmetric dimethylarginine (ADMA) on histone and non-histone proteins. It functions as a transcriptional coactivator, interacting with nuclear receptors and other transcription factors, and is involved in RNA splicing, cell cycle regulation, and signal transduction. PRMT2 is widely expressed and has been implicated in cancer, particularly breast cancer, and inflammatory responses.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Breast Cancer PRMT2 overexpression promotes cell proliferation and migration; acts as a coactivator of estrogen receptor alpha (ERα). PMID: 21947297, COSMIC
Prostate Cancer PRMT2 enhances androgen receptor (AR) transcriptional activity, contributing to tumor growth. PMID: 21947297
Inflammatory Diseases PRMT2 regulates NF-κB signaling and cytokine expression, influencing inflammation. PMID: 21947297

Mutations & Variants

Hotspot Mutations
Variant Type Frequency Functional Description
c.1132C>T (p.Arg378Trp) Missense <0.01% Unknown functional effect
c.154G>A (p.Gly52Arg) Missense <0.01% Potential loss of methyltransferase activity
Mutation functional classification

Loss of Function (LOF)

Missense mutations in the catalytic domain (e.g., p.Gly52Arg) may reduce methyltransferase activity.

Gain of Function (GOF)

Not well characterized; overexpression in cancer suggests potential gain-of-function via increased coactivator activity.

Dominant Negative (DN)

No dominant-negative mutations reported.

Pathways

• Arginine and proline metabolism
• Estrogen signaling pathway
• Androgen receptor signaling pathway
• NF-kappa B signaling pathway

Protein Summary

PRMT2 is a 433-amino acid protein containing a conserved methyltransferase domain and an SH3 domain. It catalyzes the transfer of methyl groups from S-adenosylmethionine to arginine residues, producing asymmetric dimethylarginine. PRMT2 interacts with nuclear receptors (e.g., ERα, AR) and transcription factors to modulate gene expression. It also participates in pre-mRNA splicing by methylating splicing factors. The protein is predominantly nuclear but can shuttle to the cytoplasm.
