Description
OLIG1 encodes a basic helix-loop-helix (bHLH) transcription factor essential for the specification and differentiation of oligodendrocytes in the central nervous system. It regulates genes involved in myelination and is expressed in oligodendrocyte lineage cells. OLIG1 is also implicated in neural development and certain brain tumors, particularly gliomas.
Disease Associations
| Disease category | Pathophysiological mechanism | Genomic evidence |
|---|---|---|
| Glioma | OLIG1 expression is frequently upregulated in gliomas, contributing to tumor cell proliferation and differentiation arrest. | PMID: 17634366 |
| Oligodendroglioma | OLIG1 is a diagnostic marker for oligodendroglial tumors, with high expression correlating with oligodendrocyte lineage. | PMID: 11526406 |
| Multiple sclerosis | OLIG1+ oligodendrocyte precursor cells are present in demyelinated lesions but fail to mature, suggesting impaired differentiation. | PMID: 12130774 |

Protein Summary
OLIG1 is a 303-amino acid protein containing a bHLH domain that mediates DNA binding and dimerization. It forms heterodimers with other bHLH factors to regulate transcription of genes critical for oligodendrocyte maturation and myelin production. The protein localizes to the nucleus and is highly expressed in developing and adult white matter.
