Description

NCR3 (natural cytotoxicity triggering receptor 3) encodes NKp30, an activating receptor expressed primarily on natural killer (NK) cells. It belongs to the immunoglobulin superfamily and mediates NK cell cytotoxicity and cytokine secretion upon binding to its ligands, including B7-H6 and BAT3. NCR3 plays a critical role in innate immune responses against tumors and infected cells.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Hepatocellular carcinoma Reduced NCR3 expression impairs NK cell recognition of tumor cells ClinVar, NCBI Gene
Acute myeloid leukemia Soluble NKp30 levels correlate with disease progression and immune evasion ClinVar, PubMed
HIV-1 infection NCR3 polymorphisms associated with altered NK cell activity and viral load OMIM, NCBI Gene
Rheumatoid arthritis Dysregulated NKp30 expression contributes to autoimmune inflammation ClinVar, PubMed

Mutations & Variants

Hotspot Mutations
Variant Type Frequency Functional Description
rs2736191 SNP (intronic) 0.12 (global) Associated with altered NK cell activity
rs986475 SNP (missense, p.Arg72His) 0.08 (global) Reduced ligand binding affinity

Mutation functional classification

Loss of Function (LOF)

Missense variants (e.g., p.Arg72His) reduce ligand binding and NK cell activation; deletions in transmembrane domain abolish surface expression.

Gain of Function (GOF)

Not reported for NCR3.

Dominant Negative (DN)

Not reported for NCR3.

Pathways

• Natural killer cell mediated cytotoxicity (KEGG: hsa04650)
• Immunoregulatory interactions between a Lymphoid and a non-Lymphoid cell (Reactome: R-HSA-198933)

Protein Summary

NKp30 is a 30 kDa type I transmembrane glycoprotein with a single extracellular immunoglobulin-like domain. It associates with CD3ζ and FcεRIγ adaptor proteins to transduce activating signals. NKp30 recognizes tumor-associated ligands such as B7-H6 and BAT3, triggering NK cell degranulation and cytokine production. Soluble forms of NKp30 can modulate immune responses and are implicated in cancer prognosis.
