Description

The MVP gene encodes the major vault protein, the main component of the vault complex, a large ribonucleoprotein particle involved in nucleocytoplasmic transport, multidrug resistance, and cellular signaling. MVP is overexpressed in many cancers and associated with chemoresistance.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Multidrug-resistant cancer MVP overexpression increases drug efflux via vault complex, reducing intracellular drug accumulation ClinVar, COSMIC

Protein Summary

Major vault protein (MVP) is a 110 kDa protein that forms the core of the vault complex, the largest known ribonucleoprotein particle. MVP self-assembles into a barrel-shaped structure that mediates nucleocytoplasmic transport, particularly of drugs and xenobiotics. Overexpression of MVP is a hallmark of multidrug resistance in cancer cells, where it sequesters chemotherapeutic agents away from their targets. MVP also interacts with signaling molecules such as PTEN and Src, influencing cell survival and proliferation.
