Description

MOB1A encodes a member of the MOB family of proteins that function as co-activators of the Hippo signaling pathway. The protein interacts with and activates serine/threonine kinases STK3 (MST2) and STK4 (MST1), leading to phosphorylation and activation of LATS1/2 kinases, which in turn phosphorylate and inhibit YAP1 and WWTR1 (TAZ) transcriptional co-activators. This pathway regulates organ size, cell proliferation, and apoptosis. MOB1A is ubiquitously expressed and plays a critical role in tumor suppression.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Hepatocellular carcinoma Loss of MOB1A expression leads to YAP/TAZ nuclear translocation and uncontrolled proliferation PMID: 25652263
Colorectal cancer Reduced MOB1A levels correlate with poor prognosis and activation of Hippo pathway targets PMID: 29162811
Breast cancer MOB1A downregulation promotes metastasis via YAP1 activation PMID: 30356049

Mutations & Variants

Hotspot Mutations
Variant Type Frequency Functional Description
c.205C>T (p.Arg69Trp) Missense <0.1% Impaired binding to STK3/STK4, reduced Hippo signaling
c.346G>A (p.Glu116Lys) Missense <0.1% Loss of kinase activation function
c.1A>G (p.Met1Val) Start loss Rare Complete loss of protein expression
Mutation functional classification

Loss of Function (LOF)

Missense mutations (e.g., p.Arg69Trp) disrupt interaction with MST kinases, impairing LATS1/2 activation and leading to YAP/TAZ nuclear accumulation.

Gain of Function (GOF)

No gain-of-function mutations reported in MOB1A.

Dominant Negative (DN)

No dominant-negative mutations described for MOB1A.

Pathways

• Hippo signaling pathway (KEGG: hsa04390)
• Regulation of organ size (Reactome: R-HSA-2028269)
• YAP/TAZ regulation (Reactome: R-HSA-8939211)

Protein Summary

MOB1A is a 216-amino-acid protein (25 kDa) that contains a conserved MOB1/phocein domain. It functions as a scaffold and activator of the Hippo kinase cascade. The protein binds to MST1/2 via its N-terminal region and to LATS1/2 via its C-terminal region, facilitating phosphorylation events. MOB1A is predominantly cytoplasmic but can shuttle to the nucleus upon pathway activation. Post-translational modifications include phosphorylation at Ser146 by MST1/2, which enhances its activity.
