Description
MARK3 is a member of the AMPK-related kinase family that phosphorylates microtubule-associated proteins (MAPs) such as MAPT/tau, regulating microtubule stability and cell polarity. It also acts as a scaffold in signaling pathways, including the Hippo pathway, and is involved in cell cycle control and apoptosis.
Disease Associations
| Disease category | Pathophysiological mechanism | Genomic evidence |
|---|---|---|
| Cancer (various) | MARK3 dysregulation alters cell polarity and proliferation; loss of function promotes tumorigenesis | COSMIC, ClinVar |
| Alzheimer Disease | MARK3 phosphorylates tau at sites associated with neurofibrillary tangles | UniProt, NCBI |
| Hepatocellular Carcinoma | MARK3 downregulation correlates with poor prognosis and metastasis | NCBI, COSMIC |

Protein Summary
MARK3 is a 753-amino acid serine/threonine kinase with an N-terminal kinase domain, a ubiquitin-associated domain, and a C-terminal KA1 domain. It phosphorylates tau and other MAPs, regulating microtubule dynamics and cell polarity. It also interacts with STRAD and MO25 to form a complex that activates LKB1, linking energy sensing to cell growth.
