Description

MAGEA2B is a member of the melanoma-associated antigen (MAGE) gene family, which encodes cancer-testis antigens. These proteins are normally expressed only in testis and placenta but are aberrantly activated in various cancers, making them targets for cancer immunotherapy. MAGEA2B is located on chromosome Xq28 and shares high sequence similarity with other MAGE-A genes.

Disease Associations

Melanoma Overexpression of MAGEA2B as a cancer-testis antigen; recognized by cytotoxic T lymphocytes PMID: 7539677
Non-small cell lung cancer Aberrant expression associated with tumor progression and immune evasion PMID: 14559841
Breast cancer MAGEA2B expression correlates with poor prognosis and hormone receptor negativity PMID: 15604238

Mutation functional classification

Loss of Function (LOF)

No documented loss-of-function mutations in MAGEA2B.

Gain of Function (GOF)

Overexpression in tumors suggests potential gain-of-function role in immune evasion.

Dominant Negative (DN)

Not reported.

Pathways

Not assigned to any canonical pathway in Reactome or KEGG.

Protein Summary

MAGEA2B encodes a protein of 314 amino acids (UniProt P43365) belonging to the MAGE family. It contains a conserved MAGE homology domain (MHD) and is localized in the cytoplasm and nucleus. The protein is involved in protein-protein interactions and may modulate transcriptional regulation. As a cancer-testis antigen, it is a target for cancer vaccines and adoptive T-cell therapy.
