Description

LAMTOR5 encodes a component of the Ragulator complex, which is a pentameric complex that anchors the Rag GTPases to the lysosomal membrane and is essential for amino acid-dependent activation of mTORC1. The protein also functions as a scaffold in the MAPK signaling cascade and has been implicated in cell growth, proliferation, and survival. LAMTOR5 is also known as hepatitis B virus X-interacting protein (HBXIP) due to its interaction with the HBx protein.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Hepatocellular carcinoma LAMTOR5 (HBXIP) interacts with HBx protein, promoting cell proliferation and inhibiting apoptosis; overexpression correlates with poor prognosis. PMID: 15601830, PMID: 21516111
Breast cancer LAMTOR5 overexpression enhances mTORC1 signaling and cell growth; acts as an oncogene. PMID: 23327922, PMID: 26030139
Gastric cancer Upregulation of LAMTOR5 promotes tumorigenesis via mTORC1 activation. PMID: 27323871
Colorectal cancer LAMTOR5 overexpression linked to increased proliferation and metastasis. PMID: 28708997

Protein Summary

LAMTOR5 is a 91-amino acid protein that localizes to the lysosomal membrane as part of the Ragulator complex. It is essential for the recruitment and activation of mTORC1 in response to amino acids. Additionally, it serves as a scaffold for the MAPK pathway, facilitating signal transduction. Overexpression of LAMTOR5 is oncogenic in several cancers, and it interacts with the hepatitis B virus X protein to promote hepatocellular carcinoma.
