Description
KLF10 (Kruppel Like Factor 10), also known as TIEG1 (TGF-beta Inducible Early Growth Response 1), is a transcription factor that belongs to the Sp/KLF family. It contains three C2H2-type zinc finger domains and is induced by TGF-beta, BMP, and other growth factors. KLF10 regulates cell proliferation, differentiation, and apoptosis, and plays a role in bone development, immune response, and tumor suppression.
Disease Associations
| Disease category | Pathophysiological mechanism | Genomic evidence |
|---|---|---|
| Pancreatic cancer | Loss of KLF10 expression leads to increased cell proliferation and reduced apoptosis via TGF-beta signaling dysregulation | COSMIC, ClinVar |
| Breast cancer | KLF10 acts as a tumor suppressor; promoter methylation and reduced expression are associated with poor prognosis | NCBI Gene, ClinVar |
| Osteoporosis | KLF10 regulates osteoblast differentiation; knockout mice show reduced bone mass | OMIM, NCBI Gene |
| Corneal dystrophy | Mutations in KLF10 are associated with corneal endothelial dystrophy | ClinVar, OMIM |

Mutation functional classification
Loss of Function (LOF)
Nonsense and frameshift mutations leading to truncated protein or nonsense-mediated decay; reduced transcriptional activity.
Gain of Function (GOF)
Not well documented; rare missense variants may enhance stability but no confirmed gain-of-function reported.
Dominant Negative (DN)
Not reported for KLF10.

Pathways
• TGF-beta signaling pathway (Reactome: R-HSA-2173789)
• Transcriptional regulation by KLF10 (Reactome: R-HSA-9616222)
• BMP signaling pathway (KEGG: hsa04350)
Protein Summary
KLF10 encodes a 480-amino acid protein with three C2H2-type zinc finger domains at the C-terminus. It functions as a transcriptional repressor or activator depending on context. The protein is induced by TGF-beta and mediates growth inhibition by upregulating p21 and downregulating c-Myc. It also interacts with SMAD proteins to modulate TGF-beta signaling. Post-translational modifications include phosphorylation and ubiquitination.
