Description

ID1 encodes a helix-loop-helix (HLH) protein that lacks a basic DNA-binding domain. It functions as a dominant-negative regulator of basic HLH transcription factors by forming heterodimers, thereby inhibiting DNA binding and transcriptional activation. ID1 plays a key role in cell growth, differentiation, senescence, and angiogenesis. Overexpression is associated with various cancers and poor prognosis.

Protein Summary

ID1 is a 148-amino acid protein (16 kDa) belonging to the inhibitor of DNA binding family. It contains a helix-loop-helix domain but lacks a basic region, preventing direct DNA binding. ID1 heterodimerizes with E proteins (e.g., E12, E47) to sequester them from binding to E-box sequences, thereby inhibiting transcription of genes involved in differentiation. ID1 is highly expressed in embryonic tissues and certain cancers, where it promotes proliferation, angiogenesis, and metastasis.
