Description
GRAP2 (GRB2-related adaptor protein 2) encodes a member of the GRB2 family of adaptor proteins. It contains an SH2 domain flanked by two SH3 domains. GRAP2 is predominantly expressed in hematopoietic cells and plays a critical role in T-cell receptor (TCR) signaling by linking the TCR to downstream signaling pathways, including the Ras-MAPK cascade. It interacts with proteins such as LAT, SLP-76, and PLC-gamma1.

Mutation functional classification
Loss of Function (LOF)
Missense mutations in the SH2 domain (e.g., p.Arg76Trp) disrupt phosphotyrosine binding, impairing TCR signaling.
Gain of Function (GOF)
No gain-of-function mutations reported in GRAP2.
Dominant Negative (DN)
Not described for GRAP2.

Protein Summary
GRAP2 is a 330-amino-acid adaptor protein with a molecular weight of approximately 37 kDa. It contains an N-terminal SH3 domain, a central SH2 domain, and a C-terminal SH3 domain. The SH2 domain binds phosphorylated tyrosine residues on activated receptors, while the SH3 domains interact with proline-rich motifs in downstream effectors. GRAP2 is essential for linking the T-cell receptor to the Ras-MAPK pathway, thereby regulating T-cell activation, proliferation, and differentiation.
