Description

FBXO32 (F-box protein 32), also known as Atrogin-1 or MAFbx, encodes a member of the F-box protein family that functions as the substrate-recognition component of the SCF (SKP1-CUL1-F-box) ubiquitin ligase complex. It is highly expressed in skeletal and cardiac muscle and plays a critical role in muscle atrophy by targeting proteins such as MyoD, eIF3-f, and calcineurin for ubiquitin-mediated degradation. FBXO32 is induced during catabolic states (e.g., fasting, denervation, cancer cachexia) and is a key mediator of muscle wasting.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Muscle atrophy (cachexia, sarcopenia) Upregulation of FBXO32 promotes ubiquitination and degradation of muscle structural proteins and transcription factors, leading to proteolysis. PMID: 11557764, PMID: 11557765
Cardiac atrophy / heart failure FBXO32 is induced in pressure-overload and fasting models; targets calcineurin and sarcomeric proteins, contributing to cardiomyocyte loss. PMID: 15247260, PMID: 17921331
Cancer cachexia Tumor-derived factors (e.g., TNF-α, IL-6) upregulate FBXO32 via NF-κB and FoxO pathways, accelerating skeletal muscle wasting. PMID: 15181004, PMID: 21911473
Type 2 diabetes / insulin resistance FBXO32 overexpression in muscle correlates with insulin resistance; degrades IRS-1, impairing insulin signaling. PMID: 19029981
Muscle dystrophy (Duchenne) FBXO32 is elevated in dystrophic muscle; contributes to proteasomal degradation of dystrophin-associated proteins. PMID: 16917933

Mutation functional classification

Loss of Function (LOF)

Nonsense and frameshift mutations (e.g., p.Arg524*) lead to truncated protein lacking F-box domain, impairing SCF complex assembly and ubiquitin ligase activity.

Gain of Function (GOF)

No confirmed gain-of-function mutations reported in FBXO32.

Dominant Negative (DN)

Missense mutations in the F-box domain (e.g., p.Arg339Trp) may produce a dominant-negative effect by competing with wild-type FBXO32 for substrate binding without functional ubiquitination.

Pathways

• SCF ubiquitin ligase complex pathway
• FoxO signaling pathway (induction of FBXO32 during atrophy)
• NF-κB signaling pathway (cachexia-induced FBXO32)
• IGF-1/PI3K/Akt pathway (FBXO32 repression by Akt)
• Ubiquitin-proteasome pathway (muscle proteolysis)

Protein Summary

FBXO32 (Atrogin-1) is a 355-amino-acid F-box protein that serves as the substrate recognition subunit of the SCF ubiquitin ligase complex. It contains an N-terminal F-box domain (required for SKP1 binding) and a C-terminal substrate-binding domain. The protein is predominantly expressed in skeletal and cardiac muscle and is rapidly induced under catabolic conditions. Its targets include MyoD (myogenic transcription factor), eIF3-f (translation initiation factor), and calcineurin (phosphatase), whose ubiquitination and degradation promote muscle atrophy. FBXO32 is a central node in the signaling pathways that control muscle mass and is a potential therapeutic target for cachexia and sarcopenia.
