Description

ELF3 (E74 Like ETS Transcription Factor 3) is a member of the ETS family of transcription factors. It is specifically expressed in epithelial cells and plays a critical role in regulating genes involved in epithelial cell differentiation, proliferation, and barrier function. ELF3 binds to DNA sequences containing the core consensus motif 5'-GGAA-3' and acts as both a transcriptional activator and repressor. It is implicated in development of the mammary gland, prostate, lung, and intestine, and its dysregulation is associated with various cancers, including breast, prostate, and colorectal cancer.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Breast cancer ELF3 overexpression promotes cell proliferation and invasion; acts as an oncogene in luminal subtypes. ClinVar, COSMIC, PMID: 25691885
Prostate cancer ELF3 is frequently amplified and overexpressed, driving androgen receptor signaling and tumor progression. COSMIC, PMID: 23382250
Colorectal cancer Loss of ELF3 expression correlates with poor prognosis; may function as a tumor suppressor in some contexts. ClinVar, PMID: 27562873
Lung adenocarcinoma ELF3 mutations and altered expression contribute to epithelial-mesenchymal transition and metastasis. COSMIC, PMID: 29348524

Mutations & Variants

Hotspot Mutations
Variant Type Frequency Functional Description
p.Glu81Lys Missense 0.2% Alters DNA-binding affinity; potential gain-of-function
p.Arg232* Nonsense 0.1% Truncation; loss-of-function
p.Ser207Leu Missense 0.3% Reduced transcriptional activity
Amplification Copy number gain 5-10% in prostate cancer Overexpression; oncogenic
Mutation functional classification

Loss of Function (LOF)

Nonsense and frameshift mutations (e.g., p.Arg232*) lead to truncated protein lacking transactivation domain, reducing target gene activation.

Gain of Function (GOF)

Missense mutations in the ETS domain (e.g., p.Glu81Lys) can enhance DNA binding and transcriptional activity, promoting oncogenic signaling.

Dominant Negative (DN)

Some missense mutations in the DNA-binding domain may produce proteins that compete with wild-type ELF3, inhibiting normal function.

Pathways

ETS family transcription factor pathway
Epithelial cell signaling in cancer
Androgen receptor signaling pathway
EGFR signaling pathway

Protein Summary

The ELF3 protein (UniProt P78545) is a 371-amino acid transcription factor containing a conserved ETS DNA-binding domain. It localizes to the nucleus and regulates expression of genes involved in epithelial differentiation, cell cycle, and apoptosis. Post-translational modifications include phosphorylation, which modulates its activity. ELF3 interacts with co-regulators such as p300 and HDAC1 to fine-tune transcriptional output.
