Description
DUSP26 (dual specificity phosphatase 26) encodes a member of the dual specificity protein phosphatase subfamily. The encoded protein dephosphorylates phosphotyrosine and phosphoserine/threonine residues, negatively regulating MAP kinase signaling. It is involved in cell proliferation, differentiation, and apoptosis. DUSP26 has been implicated in several cancers, including neuroblastoma and thyroid cancer.
Disease Associations
| Disease category | Pathophysiological mechanism | Genomic evidence |
|---|---|---|
| Neuroblastoma | DUSP26 dephosphorylates p53, reducing its tumor suppressor activity; overexpression correlates with poor prognosis | PMID: 19903892 |
| Thyroid cancer | DUSP26 acts as a tumor suppressor by dephosphorylating ERK; loss of expression promotes tumorigenesis | PMID: 20628086 |
| Breast cancer | DUSP26 overexpression promotes cell proliferation via MAPK pathway modulation | PMID: 23382219 |

Protein Summary
DUSP26 is a 211-amino acid dual specificity phosphatase that dephosphorylates both phosphotyrosine and phosphoserine/threonine residues. It preferentially inactivates p38 and JNK MAP kinases, and also dephosphorylates p53, modulating its stability and transcriptional activity. The protein is localized in the cytoplasm and nucleus, and its expression is highest in testis and brain.
