Description

DUSP21 (dual specificity phosphatase 21) is a protein-coding gene located on chromosome 1p36.22. It encodes a member of the dual specificity phosphatase (DUSP) family, which dephosphorylates both phosphotyrosine and phosphoserine/threonine residues. DUSP21 is known to inactivate mitogen-activated protein kinases (MAPKs) such as ERK and p38, thereby regulating cell proliferation, differentiation, and stress responses. Expression is enriched in testis and certain cancer cell lines.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Cancer (general) DUSP21 may modulate MAPK signaling, influencing tumor growth and survival Limited; expression changes observed in some cancers (COSMIC)
Male infertility Potential role in spermatogenesis via MAPK regulation Hypothetical based on testis expression

Protein Summary

DUSP21 encodes a 176-amino acid dual specificity phosphatase with a conserved catalytic domain. It specifically dephosphorylates phosphothreonine and phosphotyrosine residues on MAP kinases, particularly ERK and p38. The protein is predominantly cytoplasmic and is implicated in negative regulation of cell growth and stress responses.
