Description
DNAJB6 encodes a member of the DnaJ/Hsp40 family of co-chaperones, which regulate Hsp70 chaperone activity and are critical for protein folding, degradation, and prevention of aggregation. The protein contains a conserved J-domain that mediates interaction with Hsp70s. Alternative splicing produces two major isoforms: the nuclear/cytoplasmic isoform (DNAJB6a) and the nuclear isoform (DNAJB6b). Mutations in DNAJB6 cause autosomal dominant limb-girdle muscular dystrophy type D1 (LGMDD1), characterized by progressive proximal muscle weakness and myofibrillar pathology with protein aggregates.
Disease Associations
| Disease category | Pathophysiological mechanism | Genomic evidence |
|---|---|---|
| Limb-girdle muscular dystrophy type D1 (LGMDD1) | Dominant-negative mutations impair chaperone function, leading to aggregation of Z-disk proteins and myofibrillar disorganization. | ClinVar, OMIM #603511 |
| Myofibrillar myopathy | Similar mechanism as LGMDD1; mutations in DNAJB6 cause protein aggregation and myofibrillar degeneration. | ClinVar, OMIM #611332 |
| Huntington disease (modifier) | DNAJB6 suppresses polyglutamine aggregation in cellular models; reduced expression may exacerbate pathology. | NCBI Gene, PubMed studies |
| Cancer (potential role) | Altered expression in various cancers; may influence tumor progression via chaperone activity. | COSMIC, NCBI Gene |

Mutations & Variants
Hotspot Mutations
| Variant | Type | Frequency | Functional Description |
|---|---|---|---|
| c.277C>T (p.Pro93Ser) | Missense | Reported in LGMDD1 families | Dominant-negative; disrupts J-domain function |
| c.278C>T (p.Pro93Leu) | Missense | Reported in LGMDD1 families | Dominant-negative; similar effect |
| c.335A>G (p.Asn112Ser) | Missense | Rare | Likely pathogenic; impairs chaperone activity |
| c.346G>A (p.Gly116Arg) | Missense | Reported in LGMDD1 families | Dominant-negative; causes protein aggregation |
Mutation functional classification
Loss of Function (LOF)
Not clearly established; heterozygous mutations do not cause haploinsufficiency.
Gain of Function (GOF)
Not reported.
Dominant Negative (DN)
Yes. LGMDD1 mutations produce a toxic gain-of-function by interfering with wild-type chaperone activity, leading to protein aggregation.

Protein Summary
DNAJB6 is a 36 kDa co-chaperone with a conserved N-terminal J-domain, a glycine/phenylalanine-rich region, and a C-terminal domain. It binds to Hsp70 via its J-domain, stimulating ATPase activity and facilitating substrate transfer. DNAJB6 is particularly important in muscle cells, where it prevents aggregation of Z-disk proteins such as filamin C and myotilin. Mutations in the J-domain cause LGMDD1 by a dominant-negative mechanism, leading to myofibrillar disorganization and inclusion bodies. The protein is widely expressed, with highest levels in testis and skeletal muscle.
