Description
DDX1 encodes a member of the DEAD-box helicase family, characterized by the conserved motif Asp-Glu-Ala-Asp (DEAD). This RNA helicase is involved in multiple aspects of RNA metabolism, including pre-mRNA splicing, ribosome biogenesis, RNA transport, and translation initiation. DDX1 also participates in DNA repair and transcriptional regulation. Overexpression and amplification of DDX1 are observed in several cancers, particularly neuroblastoma and breast cancer, where it may contribute to tumorigenesis and chemoresistance.
Disease Associations
| Disease category | Pathophysiological mechanism | Genomic evidence |
|---|---|---|
| Neuroblastoma | Amplification of DDX1 at 2p24.3 is frequently co-amplified with MYCN, enhancing cell proliferation and survival. | COSMIC, ClinVar |
| Breast Cancer | DDX1 overexpression correlates with poor prognosis and may promote metastasis via RNA processing alterations. | COSMIC, NCBI |
| Retinoblastoma | DDX1 interacts with RB1 protein; altered expression may disrupt cell cycle control. | UniProt, OMIM |
| Ovarian Cancer | DDX1 upregulation is associated with chemoresistance and tumor progression. | COSMIC |

Mutation functional classification
Loss of Function (LOF)
Missense mutations in helicase domain (e.g., p.Arg515Trp) may impair ATPase or RNA unwinding activity.
Gain of Function (GOF)
Gene amplification leading to overexpression is considered a gain-of-function event in neuroblastoma and breast cancer.
Dominant Negative (DN)
No confirmed dominant-negative mutations reported.

Protein Summary
DDX1 is a 740-amino acid DEAD-box RNA helicase that localizes to both nucleus and cytoplasm. It contains a conserved helicase core with ATP-binding and RNA unwinding domains, plus an N-terminal domain for protein-protein interactions. DDX1 facilitates RNA secondary structure remodeling during splicing, translation, and ribosome assembly. It also interacts with the retinoblastoma protein (RB1) and is implicated in DNA double-strand break repair via homologous recombination. In cancer, DDX1 amplification drives oncogenic RNA processing and stress adaptation.
