Description

CRISP3 encodes a cysteine-rich secretory protein (CRISP) family member predominantly expressed in the male reproductive tract, salivary glands, and neutrophils. The protein is involved in host defense, sperm maturation, and modulation of ion channel activity. CRISP3 is upregulated in prostate cancer and other inflammatory conditions.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Prostate cancer Overexpression of CRISP3 in prostate epithelium correlates with tumor aggressiveness and androgen independence PMID: 15604206; NCBI Gene
Salivary gland tumors CRISP3 is a marker for acinic cell carcinoma and adenoid cystic carcinoma PMID: 18697828; UniProt
Male infertility Altered CRISP3 levels in seminal plasma may affect sperm function PMID: 11532963; OMIM

Loss of Function (LOF)

No confirmed loss-of-function variants reported in CRISP3.

Gain of Function (GOF)

Overexpression in prostate cancer suggests potential gain-of-function role in tumor progression.

Dominant Negative (DN)

No dominant-negative mutations described.

Protein Summary

CRISP3 is a 245-amino acid secreted glycoprotein containing a CAP (cysteine-rich secretory protein, antigen 5, and pathogenesis-related 1) domain and a C-terminal CRISP domain. It is stored in neutrophil-specific granules and released upon activation. The protein modulates calcium and potassium ion channels, contributing to sperm capacitation and antimicrobial defense.
