Description

CPA2 (Carboxypeptidase A2) is a protein-coding gene located on chromosome 7q32.2. It encodes a pancreatic metallocarboxypeptidase that cleaves C-terminal aromatic and aliphatic amino acids from dietary proteins, playing a key role in digestion. The enzyme is secreted as a zymogen and activated by trypsin.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Pancreatitis Impaired CPA2 activity may contribute to autodigestion and inflammation of the pancreas. PMID: 12368274
Pancreatic cancer Altered CPA2 expression has been observed in pancreatic ductal adenocarcinoma. PMID: 25691885

Protein Summary

Carboxypeptidase A2 is a 417-amino-acid protein (UniProt P48052) synthesized as a preproenzyme in pancreatic acinar cells. After signal peptide cleavage, the proenzyme is activated by trypsin. The mature enzyme contains one zinc ion per subunit and specifically hydrolyzes C-terminal aromatic (Phe, Tyr, Trp) and aliphatic (Leu, Ile, Met) residues. It is a key component of the pancreatic digestive enzyme cocktail.
